Delayed + Sustained Release

Delaying the release of a drug may be required to protect the sensitive mucosa of the stomach from an irritant drug, the drug from gastric acid, or to deliver the drug to a specific site in the distal portion of the gastro-intestinal (GI) tract. In certain cases a delay in onset of release is followed by an immediate release of a drug at some point in the gastro-intestinal tract or it is followed by an extended release of the drug throughout the distal portion of the GI tract. Delayed followed by an extended release of a drug may be beneficial for the delivery of topical medications throughout the colon (irritable bowel syndrome), colo-rectal cancer treatment, or chronotherapeutics. Such a release profile can be achieved through coating of multi-particulates or hydrophilic matrix tablets with pH-dependent coating polymers, or coating with combinations of pH dependent coating polymers and extended release barrier membrane systems.
